玉屏风散对肺气虚型变应性鼻炎豚鼠模型IL-4、IL-12、TNF-γ表达水平及鼻腔黏病理变化的影响 |

摘要: |
目的 探讨玉屏风散对肺气虚型变应性鼻炎(allergic rhinitis,AR)豚鼠的作用机制。方法 将40只健康杂色英国种豚鼠,随机分为空白组、模型组、玉屏风组、氯雷他定组。除空白组外,其余各组均以鸡卵清蛋白(ovalbumin,OVA)与烟熏法建立肺气虚型AR模型。造模成功后,玉屏风组、氯雷他定组分别予玉屏风散[4.52 g/(kg·d)]、氯雷他定片(0.903 mg/kg)灌胃,空白组及模型组予等体积生理盐水灌胃,处理15 d后,取各组豚鼠血清及鼻腔黏膜。经HE染色法观察鼻腔黏膜细胞形态,ELISA与免疫组化法检测豚鼠血清中白细胞介素-4(interleukin-4,IL-4)、白细胞介素-12(interleukin-12,IL-12)、γ干扰素(interferon-γ,IFN-γ)等细胞因子表达水平。结果 HE染色发现,模型组细胞形态结构明显紊乱,玉屏风组、氯雷他定组细胞形态结构异常状态相对减轻。ELISA结果显示,与空白组比较,模型组、玉屏风组、氯雷他定组豚鼠血清中IL-4、IL-12表达水平升高(P<0.05),IFN-γ表达水平降低(P<0.05);与模型组比较,玉屏风组、氯雷他定组豚鼠血清IL-4与IL-12含量降低(P<0.05),氯雷他定组IFN-γ表达水平升高(P<0.05)。免疫组化染色显示,与空白组比较,模型组豚鼠鼻腔黏膜IL-4、IL-12表达明显降低(P<0.05),IFN-γ表达明显升高(P<0.01);与模型组比较,玉屏风组与氯雷他定组IL-4表达明显降低(P<0.05),氯雷他定组IL-12表达明显降低(P<0.05)。结论 玉屏风散可能通过影响IL-4、IL-12、IFN-γ水平,改变鼻腔黏膜细胞形态治疗肺气虚型AR。 |

Effects of Yupingfeng Powder on expression levels of IL-4,IL-12,and TNF-γ as well as nasal mucosa pathological changes in guinea pig model of allergic rhinitis with lung qi deficiency pattern |

Abstract: |
Objective To explore the action of mechanism of Yupingfeng Powder (YPFP) on allergic rhinitis (AR) with lung qi deficiency pattern in guinea pigs.Methods Forty healthy variegated British guinea pigs were randomized into blank group,model group,YPFP group,and loratadine group.Except for the blank group,the AR models with lung qi deficiency pattern were established in the other groups using ovalbumin (OVA) and smoke exposure.After successful modeling,the YPFP group and the loratadine group were treated with YPFP[4.52 g/(kg·d)]and loratadine tablets (0.903 mg/kg) by gavage,respectively,while the blank group and model group were given an equal volume of normal saline by gavage.After 15 days of treatment,the serum and nasal mucosa samples of guinea pigs were collected from each group.The morphology of nasal mucosa cells were observed using HE staining,and the expression levels of cytokines such as interleukin-4(IL-4),interleukin-12(IL-12),and interferon-γ(IFN-γ) in the serum of guinea pigs were measured by ELISA and immunohistochemical staining.Results HE staining showed that the model group exhibited significant morphological and structural disorders,while the morphological and structural abnormalities were relatively alleviated in the YPFP group and the loratadine group.ELISA results indicated that compared with the blank group,the expression levels of IL-4 and IL-12 in the serum of guinea pigs in the model group,YPFP group,and loratadine group increased (P<0.05),while the expression level of IFN-γ decreased (P<0.05).Compared with the model group,the serum levels of IL-4 and IL-12 in the YPFP group and loratadine group decreased (P<0.05),while the expression level of IFN-γ in the loratadine group increased (P<0.05).Immunohistochemical staining revealed that compared with the blank group,the expressions of IL-4 and IL-12 in the nasal mucosa of guinea pigs in the model group were significantly lower (P<0.05),while the expression of IFN-γ was significantly higher (P<0.01).Compared with the model group,the IL-4 expression decreased significantly in both the YPFP group and loratadine group (P<0.05),and the IL-12 expression in the loratadine group decreased significantly (P<0.05).Conclusion YPFP may treat allergic rhinitis with lung qi deficiency pattern by influencing the levels of IL-4,IL-12,and IFN-γ,and changing the morphology of nasal mucosa cells. |
