水飞蓟喷雾剂对大鼠深Ⅱ度烫伤创面修复的影响 |

摘要: |
目的 考察水飞蓟喷雾剂对大鼠深Ⅱ度烫伤创面修复作用及机制。方法 建立深Ⅱ度烫伤模型大鼠,随机分为模型组、美宝湿润烧伤膏组(24 g/kg)、水飞蓟喷雾剂小剂量组(24 g/kg)、水飞蓟喷雾剂中剂量组(48 g/kg)、水飞蓟喷雾剂大剂量组(96 g/kg)和空白组,连续给药21 d。分别在造模后第3、7、14、21天计算烫伤创面愈合率,检测血清超氧化物歧化酶(superoxide dismutase,SOD)活性和丙二醛(malondialdehyde,MDA)含量;免疫组化法检测烫伤创面碱性成纤维细胞生长因子(basic fibroblast growth factor,b-FGF)、表皮生长因子(epidermal growth factor,EGF)、转化生长因子-β(transformating growth factor betal,TGF-β)的表达。结果 与模型组相比,烫伤后第3天,水飞蓟喷雾剂小剂量组、中剂量组、大剂量组创面愈合率提高,血清SOD活性升高,MDA含量降低,且差异均具有统计学意义(P<0.05);烫伤后第7、14天,水飞蓟喷雾剂大剂量组血清SOD活性升高,MDA含量降低,差异均具有统计学意义(P<0.05);同时水飞蓟喷雾剂各剂量组均可不同程度诱导受损组织内bFGF、EGF、TGF-β的表达。结论 水飞蓟喷雾剂对大鼠深Ⅱ度烫伤创面有修复作用,可促进烫伤创面的愈合,其机制可能与抗氧化,增加b-FGF、EGF、TGF-β的表达有关。 |

Efficacy of Silymarin Spray in Treatment of Deep Ⅱ Degree Scald Wound in Rats |

Abstract: |
Objective To investigate the efficacy of silymarin spray in the treatment of deep Ⅱ degree scald wound in rats and its mechanism of action.Methods A model of deep Ⅱ degree scald wound was established in rats. The rats were randomly divided into model group, moisture burn ointment (MEBO) group (24 g/kg), low-dose silymarin spray group (24 g/kg), medium-dose silymarin spray group (48 g/kg), high-dose silymarin spray group (96 g/kg), and blank group. They were given respective treatments for 21 consecutive days. At 3, 7, 14, and 21 days after modeling, the healing rate of scald wound was calculated, and the serum levels of superoxide dismutase (SOD) and malondialdehyde (MDA) were measured; the expression of basic fibroblast growth factor (b-FGF), epidermal growth factor (EGF), and transforming growth factor beta (TGF-β) in the wound was measured by immunohistochemistry.Results Compared with the model group, at 3 days after scalding, the three silymarin spray groups had significantly higher wound healing rate and serum level of SOD and a significantly lower serum level of MDA (all P<0.05); at 7 and 14 days after scalding, the high-dose silymarin spray group had a significantly higher serum level of SOD and a significantly lower serum level of MDA (all P<0.05); the expression of b-FGF, EGF, and TGF-β in the damaged tissue was increased to varying degrees in the three silymarin spray groups (all P<0.05).Conclusion Silymarin spray has a repairing effect on deep Ⅱ degree scald wound in rats by promoting the healing of scald wound. The mechanism may be related to antioxidation and increasing the expression of b-FGF, EGF, and TGF-β. |
