摘要: |
目的观察局灶性脑缺血模型大鼠脑组织凝血酶原(prothrombin , F2) mRNA和蛋白酶激活受体一I (PrO-tease-activated receptor-1 ,PAR-1)mRNA表达变化及滋阴活血解毒方的千预作用方法将24只大鼠随机分为假手术组,模型组、滋阴活血解毒方组、阿加曲班4组,每组分1,3 d两个时相点〔制备大脑中动脉闭塞(MCAO)模型,检测各组大鼠脑组织中凝血酶原mRNA及PAR-1mRNA的表达水平、结果与假手术组比较,模型组脑组织中凝血酶原mRNA ,PAR-1mRNA表达明显增强(P<0.01) ,滋阴活血解毒方组和阿加曲班组能明显下调凝血酶原mRNA,PAR-1mRNA表达(P<0.01)〔结论局灶性脑缺血大鼠凝血酶原mRNA和PAR-1mRNA的表达增强,滋阴活血解毒方能抑制其表达,减轻凝血酶的毒性反应、 |

Abstract: |
Objective To observe the changes of Prothrombin mRNA and PAR-1 mRNA ex-pression in focal cerebral infarction in rats and intervening effects of Ziyin Huoxue Jiedu decoc-tion. Methods Twenty-four rats were randomly divided into 4 groups: the sham一(operated, model,Ziyin Huoxue Jiedu decoction and Argatroban groups. Each group was divided into two time phase point of 1st day and 3rd day. The MCAO models were reproduced. The expression of prothrombin mRNA and PAR-1 mRNA in brain tissue were detected. Results Compared with the sham-operat-ed group, the expression of prothrombin mRNA and PAR-1 mRNA in brain tissue in model group were significantly higher (P<0.01), the expression of prothrombin mRNA and PAR一1 mRNA were lowered significantly in Ziyin Huoxue Jiedu decoction group and Argatroban Injection group (P<0.01). Conclusion Ziyin Huoxue Jiedu decoction is effective[()restrain the expression of prothrom-bin mRNA and PAR-1 mRNA in brain tissue and reduce thrombin toxic injury. |
