摘要: |
目的 通过观察针刺或热灸不同刺激方式干预足三里穴区对C57BL/6野生型小鼠和TRPV1-/-基因敲除小鼠血浆中脑肠肽(ghrelin)和大脑前额叶皮质中生长激素促分泌素受体-1α(GHSR-1α)含量的改变,探讨辣椒素受体1(TRPV1)参与针、灸不同刺激方式对胃肠功能调控通路ghrelin-GHSR-1α调节的差异性。方法 24只C57BL/6野生型小鼠随机分为C57BL/6空白组、C57BL/6针刺组和C57BL/6热灸组,每组8只;24只TRPV1-/-基因敲除小鼠随机分为TRPV1-/-空白组、TRPV1-/-针刺组和TRPV1-/-热灸组,每组8只。C57BL/6空白组和TRPV1-/-空白组不予以处理;C57BL/6针刺组和TRPV1-/-针刺给予手针干预;C57BL/6热灸组和TRPV1-/-热灸组给予热灸干预。采用ELISA检测小鼠血浆中ghrelin含量和Western Blot法检测小鼠大脑前额叶皮质中TRPV1和GHSR-1α表达水平的变化。结果 C57BL/6野生型小鼠中,与C57BL/6空白组相比,C57BL/6针刺组与热灸组血浆ghrelin表达水平均显著降低(P<0.05),前额叶皮层TRPV1含量表达增加(P<0.05)、GHSR-1α降低(P<0.05),但三者在针刺组和热灸组间差异无统计学意义(P>0.05)。TRPV1-/-基因敲除小鼠中,与TRPV1-/-空白组对比,TRPV1-/-针刺组和热灸组血浆ghrelin水平和前额叶皮质GHSR-1α表达水平差异均无统计学意义(P>0.05)。结论 针刺、热灸均可通过激活TRPV1的生物活性,影响胃肠功能调控通路ghrelin-GHSR-1α的调节作用。 |

Effect of TRPV1 on ghrelin and GHSR-1α in Mice by Participating in Different Stimulation Methods of Acupuncture and Moxibustion |

Abstract: |
Objective To observe the effect of different stimulation modalities of acupuncture or pyretic moxibustion in the "Zusanli" (ST36) acupoint on changes of content of plasma ghrelin and growth hormone secretagogue receptor-1 (GHSR-1α) in the prefrontal cortex of C57BL/6 wild-type mice and TRPV1-/- knockout mice, and to explore the difference of capsaicin receptor 1 (TRPV1) involved in acupuncture and pyretic moxibustion stimulation on ghrelin-GHSR-1α regulation of gastrointestinal function pathway. Methods 24 C57BL/6 wild-type mice were randomly divided into C57BL/6 blank group, C57BL/6 acupuncture group and C57BL/6 hot pyretic moxibustion group, with 8 mice in each group. 24 TRPV1-/- knockout mice were randomly divided into TRPV1-/- blank group, TRPV1-/- acupuncture group and TRPV1-/- pyretic moxibustion group, with 8 mice in each group. C57BL/6 blank group and TRPV1-/- blank group were not treated; C57BL/6 acupuncture group and TRPV1-/- acupuncture group were given hand acupuncture intervention; C57BL/6 pyretic moxibustion group and TRPV1-/- pyretic moxibustion group were given pyretic moxibustion intervention. ELISA was used to detect the ghrelin content in mouse plasma, and Western blot was used to detect the changes of TRPV1 and GHSR-1α expression levels in mouse brain prefrontal cortex. Results In C57BL/6 wild-type mice, compared with the C57BL/6 blank group, plasma ghrelin level in C57BL/6 acupuncture group and pyretic moxibustion group was significantly decreased (P<0.05), while the content of TRPV1 in prefrontal cortex was increased (P<0.05) and GHSR-1α was decreased (P<0.05), but there was no statistically significant difference between the acupuncture group and pyretic moxibustion group (P>0.05). In TRPV1-/- knockout mice, compared with the TRPV1-/- blank group, there was no statistically significant difference in plasma ghrelin level and prefrontal cortex GHSR-1α expression level in TRPV1-/- acupuncture group and pyretic moxibustion group (P>0.05). Conclusion Both acupuncture and pyretic moxibustion can affect the regulation of gastrointestinal functional pathway ghrelin-GHSR-1α by activating the biological activity of TRPV1. |
