海金子中柽柳素-3-O-芸香糖苷和芦丁的

摘要: |
目的研究海金子70%乙醇提取物的化学成分;建立HPLC 测定海金子中柽柳素-3-O-芸香糖苷、芦丁的
含量。方法采用硅胶柱色谱进行分离纯化,利用波谱技术、色谱技术进行结构鉴定。采用Agela Technologies Promosil
C18(200 mm×4.6 mm,5 μm),流动相为甲醇-0.4%磷酸水溶液(34∶66),流速为1.0 mL/min,检测波长为254 nm,柱温为
40 ℃对不同采集时间、不同部位的海金子中的柽柳素-3-O-芸香糖苷、芦丁进行含量测定。结果分离鉴定了2 个黄
酮苷类化合物,分别为柽柳素-3-O-芸香糖苷和芦丁。柽柳素-3-O-芸香糖苷在0.254~1.145 μg 范围、芦丁在0.341~
1.533 μg 范围与其峰面积呈良好的线性关系;平均回收率:柽柳素-3-O-芸香糖苷为100.08%,RSD 为1.09%(n=6),芦
丁为99.89%,RSD 为1.41%(n=6)。结论两种化合物首次从该植物中分离得到。所建立的HPLC 简便、准确、重现性
好,结果准确可靠,可用于海金沙中柽柳素-3-O-芸香糖苷和芦丁的测定。 |

Abstract: |
Objective To investigate the chemical constituents in 70% alcohol extract of Pittosporum.
Illicioides Mak. and develop an HPLC method for determination of tamarix prime-3-Orutinoside
and rutin in Pittosporum. Illicioides Mak. Methods The chemical constituents were isolated
by column chromatography over silica gel. The Structures of the two compounds were elucidated
by spectroscopic and chromatographic analysis. Agela Technologies Promosil C18 column
(200 mm×4.6 mm, 5 μm) was used with methanol-0.4% phosphoric acid (34:66) as mobile phase.
The flow rate was 1.0 mL/min. The UV detection wavelength was 254 nm and the column temperature
was 40 ℃. According to these conditions, the contents of tamarix prime-3-O-rutinoside and
rutin in different parts of Pittosporum. Illicioides Mak cropped at different time were determined.
Results The two flavone glycosides were identified as tamarix prime-3-O-rutinoside and rutin. The
linear response range of tamarix prime -3 -O -rutinoside and rutin were 0.254 ~1.145 μg and,
0.341 ~1.533 μg, respectively. The average recoveries of these compounds were 100.08% (RSD =
1.09%) and 99.89%(RSD=1.41%), respectively. Conclusion The two kinds of compounds were isolated
from Pittosporum. Illicioides Mak for the first time. This established method in this study is
simple, reliable, reproducible and accurate for the analysis of tamarix prime-3-O-rutinoside and
rutin in Pittosporum. Illicioides Mak. |
