基于OPG/RANK/RANKL信号通路探讨中医学“从虚从瘀”论治骨质疏松症的科学内涵

摘要:
    骨质疏松症(OP)的发生发展与中医学的肝肾亏虚、血瘀病机密切相关,但其具体分子机制尚待深入阐明。骨保护素(OPG)/核因子κB受体活化因子(RANK)/核因子κB受体活化因子配体(RANKL)信号通路作为调控破骨细胞分化与骨吸收的重要分子通路,已成为当前防治OP的重要研究靶点。本文立足中医学“从虚从瘀”的辨证论治思想,以OPG/RANK/RANKL信号通路为切入点,探讨肝肾亏虚与血瘀病机在OP发病中的分子生物学基础及内在联系,旨在为中医药干预OP提供现代医学的理论依据与科学支撑,并进一步拓展中医药在防治OP领域的研究与临床应用新思路。

Scientific connotation of Chinese medicine principle of "treating osteoporosis from deficiency and blood stasis" based on the OPG/RANK/RANKL signaling pathway

Abstract:
    The occurrence and progression of osteoporosis(OP) are closely associated with the Chinese medicine(CM)pathogenesis of liver and kidney deficiency and blood stasis. However, the specific molecular mechanisms underlying remain to be further elucidated. The osteoprotegerin(OPG)/receptor activator of nuclear factor-κB(RANK)/receptor activator of nuclear factor-κB ligand(RANKL) signaling pathway, a key regulator of osteoclast differentiation and bone resorption, has emerged as a critical target for the current prevention and treatment of OP. Grounded in the CM pattern differentiation and treatment principle of "treating osteoporosis from deficiency and blood stasis", this paper explores the molecular biological basis and internal connections of liver and kidney deficiency and blood stasis in the pathogenesis of OP through the lens of the OPG/RANK/RANKL signaling pathway.The aim is to provide a theoretical and scientific foundation from modern medicine for CM-based interventions in OP, and to offer new perspectives for further research and clinical application of CM in the prevention and treatment of OP.
