摘要: |
目的 建立丹润方指纹图谱并测定没食子酸、绿原酸、鞣花酸含量,为其质量控制提供依据。方法 采用Agilent 5 TC-C18色谱柱,以乙腈-0.1%磷酸水溶液为流动相,梯度洗脱,流速1.0 mL·min-1,检测波长315 nm,柱温30 ℃;通过相似度评价,结合主成分分析及偏最小二乘法-判别分析对15批丹润方指纹图谱进行分析评价,并对3种指标成分进行定量测定。结果 建立了15批丹润方指纹图谱,共标定19个共有峰,各批次的相似度均大于0.9;没食子酸、绿原酸、鞣花酸在各自线性范围内线性关系良好;15批丹润方中没食子酸、绿原酸、鞣花酸的质量分数为0.219~1.100、0.168~0.445、0.418~3.077 mg/g。结论 建立的丹润方指纹图谱及多成分定量测定方法简单、准确、重现性较好,可用于丹润方整体质量评价。 |

Objective To establish the HPLC fingerprint of Danrun Formula and determine the content of gallic acid, chlorogenic acid, and ellagic acid so as to provide basis for its quality control. Methods Agilent 5 TC-C18 chromatographic column was used, with the mobile phase of cetonitrile-0.1% phosphoric acid aqueous solution for gradient elution at a flow rate of 1.0 mL·min-1. The detection wavelength was 315 nm, and the column temperature was 30 ℃. Through similarity evaluation, combined with principal component analysis and partial least squares-discriminant analysis, the fingerprints of 15 batches of Danrun Formula were analyzed and evaluated, and the 3 index components were quantitatively determined. Results The fingerprints of 15 batches of Danrun Formula were established with 19 common peaks, and the similarity of each batch was greater than 0.9. Gallic acid, chlorogenic acid, and ellagic acid had good linear relationships within their respective linear ranges. The mass fractions of gallic acid, chlorogenic acid, and ellagic acid in 15 batches of Danrun Formula were 0.219-1.100, 0.168-0.445, and 0.418-3.077 mg/g, respectively. Conclusion The established fingerprint and multi-component quantitative determination method of Danrun Formula are simple, accurate, and reproducible, which can be used for the overall quality evaluation of Danrun Formula. |
