基于TGF-β1/Smads信号通路研究丹荔输通汤对慢性输卵管炎性阻塞模型大鼠的影响 |

摘要: |
目的 通过TGF-β1/Smads信号通路,研究丹荔输通汤对慢性输卵管炎性阻塞模型大鼠的影响。方法 将造模后大鼠随机分为模型组、高剂量灌胃+灌肠组(14 g·kg-1)、中剂量灌胃+灌肠组(7 g·kg-1)、低剂量灌胃+灌肠组(3.5 g·kg-1)、中剂量灌胃组(7 g·kg-1)、对照组(阿奇霉素,1 mg·kg-1),另设空白组。予相应药物治疗30 d后处死全部大鼠,取输卵管组织,病理检测观察输卵管组织的组织形态变化,以RT-PCR、Western blot检测各组大鼠输卵管组织中TGF-β1、Smad2、Smad3 mRNA与蛋白的表达。结果 与模型组相比,各组大鼠输卵管肿胀、充血等表现均有所减轻,各组大鼠输卵管组织中的TGF-β1、Smad2、Smad3 mRNA与蛋白的表达均明显降低(P<0.01);高剂量灌胃+灌肠组、中剂量灌胃+灌肠组大鼠输卵管组织中的TGF-β1、Smad2、Smad3 mRNA与蛋白的表达均低于低剂量灌胃+灌肠组、中剂量灌胃组、对照组(P<0.01)。结论 丹荔输通汤能明显改善输卵管炎性阻塞模型大鼠输卵管组织炎症反应,改善纤维化水平,其可能通过下调TGF-β1、Smad2、Smad3 mRNA与蛋白表达,通过TGF-β1/Smads信号通路发挥作用。 |

Effects of Danli Shutong Decoction on chronic tubal inflammatory obstruction in rats based on TGF-β1/Smads signaling pathway |

Objective To study the effects of Danli Shutong Decoction on rats with chronic tubal inflammatory obstruction based on TGF-β1/Smads signaling pathway. Methods The modeled rats were randomly divided into model group, high-dose intragastric gavage+enema group (14 g·kg-1), middle-dose intragastric gavage+enema group (7 g·kg-1), low-dose intragastric gavage+enema group (3.5 g·kg-1), meddle-dose intragastric group (7 g·kg-1), control group (azithromycin, 1 mg·kg-1), and blank group. After 30 d of corresponding drug treatment, all rats were sacrificed, and then their fallopian tube tissues were then taken out. We carried out pathological detectionof the tissue morphological changes of fallopian tube tissue. The expression of TGF-β1, Smad2, Smad3 mRNA and the protein in fallopian tube tissues of each group was detected by RT-PCR and Western blot. Results Compared with model group, the swelling and congestion of fallopian tubes of rats in each group was reduced. The expression of TGF-β1, Smad2, Smad3 mRNA and the protein in fallopian tubes of rats in each group decreased significantly (P<0.01); the expression of TGF-β1, Smad2, Smad3 mRNA and the protein in oviduct tissue of rats in high-dose intragastric gavage+enema group and middle-dose intragastric gavage+enema group were lower than that in middle-dose intragastric gavage+enema group, low-dose intragastric gavage group and control group (P<0.01). Conclusion Danli Shutong Decoction could significantly improve the inflammatory reaction of fallopian tube tissue and the level of fibrosis in rats with fallopian tube inflammatory obstruction, which may play a role by TGF-β1/Smad2 signaling pathway through down regulating the expression of TGF-β1, Smad2, Smad3 mRNA and the protein. |
