摘要: |
目的 观察青光安颗粒剂对抗青光眼术后滤过道组织自噬相关蛋白Beclin1/微管相关蛋白轻链B(light chain 3B,LC3B)表达的影响。方法 将30只成功建立慢性高眼压模型的新西兰白兔随机平均分为5组,除空白组(A组)外,其余各组均行常规小梁切除术,阳性对照组(B组)术中放置丝裂霉素C棉片,单纯手术组(C组)仅行常规小梁切除术,青光安组(D组)术后灌胃青光安颗粒剂混悬液,阴性对照组(E组)术后灌胃蒸馏水。干预14 d后,Western blot法检测滤过道组织Beclin1、LC3B-I、LC3B-II的表达量,RT-qPCR法检测Beclin1及LC3B mRNA的相对表达量。结果 各手术组Beclin1蛋白平均表达量均显著高于A组(P<0.05),B组Beclin1蛋白平均表达量显著高于C组、D组与E组(P<0.05),D组Beclin1蛋白平均表达量高于C、E组(P<0.05);各手术组LC3B-II/LC3B-I均显著高于A组(P<0.05),B组LC3B-II/LC3B-I显著高于C、D、E组(P<0.05),D组LC3B-II/LC3B-I高于C、E组(P<0.05)。各手术组Beclin1 mRNA相对表达量均显著高于A组(P<0.05),B组Beclin1 mRNA相对表达量显著高于C组与E组(P<0.05),B组Beclin1 mRNA相对表达量与D组无明显区别(P>0.05);各手术组LC3B mRNA相对表达量均显著高于A组(P<0.05),B组LC3B mRNA相对表达量显著高于C组与E组(P<0.05),B组LC3B mRNA相对表达量与D组无明显区别(P>0.05)。结论 青光安颗粒剂能通过促进滤过道组织Beclin1蛋白及mRNA的表达,诱导瘢痕组织的自噬,维持滤过道的通畅。 |

Objective To observe the effect of Qingguang’an Granule on the expression of autophagy-related protein Beclin1/light chain 3B (LC3B) in filtration duct tissue after anti-glaucoma surgery. Methods Thirty New Zealand white rabbits successfully established a chronic high intraocular pressure model were randomly divided into 5 groups. Except for the blank group (group A), all other groups underwent routine trabeculectomy. The positive control group (group B) was placed mitomycin C cotton tablets during operation, the simple operation group (group C) only underwent conventional trabeculectomy, the Qingguang’an group (group D) was given Qingguang’an Granule suspension after surgery, the negative control group (group E) was given distilled water after operation. After 14 days of intervention, Western blot method was used to detect the expression levels of Beclin1, LC3B-I, and LC3B-II in filtration duct tissue, and the relative expression levels of Beclin1 and LC3B mRNA were detected by RT-qPCR. Results The average expression of Beclin1 protein in each operation group was significantly higher than that in group A (P<0.05), the average expression of Beclin1 protein in group B was significantly higher than that in group C, group D and group E (P<0.05), and the average expression of Beclin1 protein in group D was significantly higher than that in group C and group E (P<0.05); LC3B-II/LC3B-I in each operation group was significantly higher than that in group A (P<0.05), LC3B-II/LC3B-I in group B was higher than that in group C, group D and group E (P<0.05), LC3B-II/LC3B-I in group D was higher than that in group C and group E (P<0.05). The relative expression of Beclin1 mRNA in each operation group was significantly higher than that in group A (P<0.05), the relative expression of Beclin1 mRNA in group B was significantly higher than that in group C and group E (P<0.05), the relative expression of Beclin1 mRNA in group B was not significantly different from that in group D (P>0.05); the relative expression of LC3B mRNA in each operation group was significantly higher than that in group A (P<0.05), and the relative expression of LC3B mRNA in group B was significantly higher than that in group C and group E (P<0.05), the relative expression of LC3B mRNA in group B was not significantly different from group D (P>0.05). Conclusion Qingguang’an Granule can induce the autophagy of scar tissue by promoting the expression of Beclin1 protein and mRNA in the filtration duct tissue, and maintain the patency of the filtration duct tissue. |
