UPLC-MS/MS法同时测定杞菊地黄胶囊中7种有效成分

摘要:
    目的 建立超高效液相色谱-串联质谱(UPLC-MS/MS)同时测定杞菊地黄胶囊中莫诺苷、绿原酸、马钱苷、木犀草苷、丹皮酚、23-乙酰泽泻醇C、23-乙酰泽泻醇B的含量。方法 采用ACQUITY UPLC® HSS T3(100 mm×2.1 mm,1.8 μm)为色谱柱,乙腈-0.1%甲酸水溶液为流动相,梯度洗脱,流速为0.3 mL·min-1;采用电喷雾电离源(ESI)、多反应离子监测(MRM)模式对7种有效成分进行测定。结果 每粒杞菊地黄胶囊样品中莫诺苷含量为0.28~0.35 mg,绿原酸含量为0.08~0.11 mg,马钱苷含量为0.30~0.48 mg,木犀草苷含量为0.21~0.48 mg,丹皮酚含量为0.62~0.76 mg,23-乙酰泽泻醇C含量为0.04~0.08 mg,23-乙酰泽泻醇B含量为0.15~0.32 mg。7种有效成分相关系数为0.999 0~1.000 0;平均回收率(n=6)为97.72%~99.12%,RSD为0.61%~2.01%。结论 该方法专属性强、灵敏高、重复性好,适用于杞菊地黄胶囊中莫诺苷、绿原酸、马钱苷、木犀草苷、丹皮酚、23-乙酰泽泻醇C和23-乙酰泽泻醇B 7种有效成分的同时测定。

Simultaneous Determination of Seven Effective Components in Qiju Dihuang Capsule by UPLC-MS/MS

Abstract:
    Objective To establish an UPLC-MS/MS method for simultaneous determination of seven effective components (morroniside, chlorogenic acid, loganin, luteolin-7-O-glucoside, paeonol, 23-acetate alisol C, 23-acetate alisol B) in Qiju Dihuang Capsule. Methods The ACQUITY UPLC® HSS T3 (100 mm×2.1 mm, 1.8 μm) column was adopted with mobile phase consisted of acetonitrile-0.1%-formic acid with gradient elution at the flow rate of 0.3 mL·min-1. Electrospray ionization (ESI) and multiple reactive ion monitoring (MRM) modes were used to determine the seven active components. Results The contents of morroniside, chlorogenic acid, loganin, luteolin-7-O-glucoside, paeonol, 23-acetate alisol C, 23-acetate alisol B in the samples per capsule were 0.28~0.35, 0.08~0.11, 0.30~0.48, 0.21~0.48, 0.62~0.76, 0.04~0.08, 0.15~0.32 mg. Correlation coefficient values of the 7 active components were among 0.999 0~1.000 0; the average recovery rates were among 97.72%~99.12% (n=6), and the RSD were among 0.61%~2.01%. Conclusion The method established is specific, sensitive and reproducible, which is suitable for the determination of seven effective components (morroniside, chlorogenic acid, loganin, luteolin-7-O-glucoside, paeonol, 23-acetate alisol C, 23-acetate alisol B) in Qiju Dihuang Capsule.
