调心通督法对血管性痴呆大鼠学习记忆能力及海马区bFGF表达的影响 |

摘要: |
目的 探讨调心通督法对血管性痴呆(vascular dementia,VD)大鼠学习记忆功能及海马碱性成纤维细胞生长因子(basic fibroblast growth factor,bFGF)表达的影响。方法 SD大鼠行Morris水迷宫实验筛选出特异性大鼠后随机分出6只假手术组,其余大鼠采用双侧颈总动脉永久性结扎法(2-VO)复制VD大鼠模型,造模后再按随机数字表法分为模型组、西药对照组、调心通督法针刺组,每组6只。于造模成功后及治疗后分别采用Morris水迷宫实验观察大鼠学习记忆能力,Western Blot法检测双侧海马bFGF蛋白表达。结果 造模后,与假手术组比较,各组大鼠平均逃避潜伏期时间明显增加,且找到平台次数明显减少(P<0.01)。治疗后,与模型组相比,西药对照组、调心通督法针刺组平均逃避潜伏期明显缩短(P<0.01),穿越原平台次数增加(P<0.05);与西药对照组比较,调心通督法针刺组差异不具有统计学意义(P>0.05)。与假手术组比较,各组大鼠海马区bFGF表达均升高(P<0.01);与模型组比较,西药对照组及调心通督法针刺组海马bFGF表达均明显增高(P<0.05)。结论 调心通督法能明显改善VD大鼠学习记忆能力、上调海马bFGF表达,可能是其重要作用机制之一。 |

Objective To explore the effects of Tiaoxin Tongdu method on learning and memory function and basic fibroblast growth factor (bFGF) expression in hippocampus of vascular dementia (VD) rats. Methods SD rats were selected by Morris water maze test and then randomly divided into 6 sham operation groups. VD rat model was established by permanent ligation of bilateral common carotid artery (2-VO). The other rats were divided into a model group, a western medicine control group and Tiaoxin Tongdu method acupuncture group according to the random number table method, with 6 rats in each group. Morris water maze test was used to observe the learning and memory ability of rats after successful modeling and after treatment. Western Blot method was used to detect the expression of bFGF protein in bilateral hippocampus. Results Compared with sham operation groups, the average escape latency time of rats in each group increased significantly, and the number of platforms found decreased significantly (P<0.01). Compared with the model group, the average escape latency was significantly shortened (P<0.01) and the number of times crossing the original platform was increased (P<0.05) in the western medicine control group and the Tiaoxin Tongdu method acupuncture group. Compared with the western medicine control group, the difference of the Tiaoxin Tongdu method acupuncture group was not statistically significant (P>0.05). Compared with the sham operation groups, the expression of bFGF in hippocampus of each group was increased (P<0.01). Compared with the model group, the expression of bFGF in hippocampus of the western medicine control group and the Tiaoxin Tongdu method acupuncture group increased significantly (P<0.05). Conclusion Tiaoxin Tongdu method can significantly improve the learning and memory ability of VD rats, and up-regulation of bFGF expression in hippocampus may be one of its important mechanisms. |
