清霾饮防治PM2.5致大鼠肺损伤的研究 |

摘要: |
目的 研究清霾饮对大气细颗粒物PM2.5所致大鼠肺损伤的防治作用。方法 将健康成年SD大鼠50只随机分为空白组、PM2.5染毒组、清霾饮低剂量干预组、清霾饮中剂量干预组、清霾饮高剂量干预组。采集空气中细颗粒物制成染毒造模所需浓度为37.5 mg/mL的颗粒物混悬液处理大鼠建立肺损伤模型,并给予不同剂量清霾饮进行干预。最后一次染毒48 h后,收集腹主动脉血及右侧肺组织支气管肺泡灌洗液,测定血清中酸性磷酸酶(acid phosphatase,ACP)、乳酸脱氢酶(lactic dehydrogenase,LDH)、谷胱甘肽过氧化物酶(glutathion peroxidase,GSH-Px)活性和支气管肺泡灌洗液(BALF)中ACP、LDH活性,并观察左侧肺组织病理学改变。结果 与空白组相比,PM2.5染毒组BALF及血清中ACP、LDH的活性均明显升高;血清中GSH-Px活性降低,差异具有统计学意义(P<0.01)。与PM2.5染毒组相比,清霾饮低剂量干预组各测定指标差异均无统计学意义(P>0.05);而清霾饮中、高剂量干预组大鼠BALF及血清中ACP、LDH活性明显降低;血清中GSH-Px活性升高,差异具有统计学意义(P<0.05)。结论 清霾饮能拮抗PM2.5对肺组织所产生的炎性及氧化损伤,对肺组织有保护作用。 |

Qingmai Decoction in the Prevention and Treatment of Fine Particulate Matter-Induced Pulmonary Impairment in Rats |

Abstract: |
Objective To investigate the significance of Qingmai Decoction in the prevention and treatment of fine particulate matter (PM2.5)-induced pulmonary impairment in rats. Methods Fifty healthy adult Sprague-Dawley rats were randomly divided into blank group, PM2.5 exposure group, low-dose Qingmai Decoction group, medium-dose Qingmai Decoction group, and high-dose Qingmai Decoction group. Fine particles in the air were collected and made into a liquid suspension with a concentration of 37.5 mg/ml, which was used to treat rats to establish a model of pulmonary impairment. Rats were given different doses of Qingmai Decoction for intervention. At 48 hours after the last exposure, abdominal aortic blood and bronchoalveolar lavage fluid (BALF) of the right lung were collected to determine the activity of acid phosphatase (ACP) and lactic dehydrogenase (LDH) in serum and BALF as well as the activity of glutathione peroxidase (GSH-Px) in serum. Pathological section of the right lung lobe was evaluated. Results Compared with the blank group, the PM2.5 exposure group had significantly higher activity of ACP and LDH in BALF and serum and significantly lower activity of GSH-Px in serum (P<0.01). There were no significant differences in any indices between the PM2.5 exposure group and low-dose Qingmai Decoction group (P>0.05). Compared with the PM2.5 exposure group, the medium-and high-dose Qingmai Decoction groups had significantly lower activity of ACP and LDH in BALF and serum and significantly higher activity of GSH-Px in serum (P<0.05). Conclusion Qingmai Decoction can protect the lung tissue by antagonizing the PM2.5-induced inflammatory and oxidative damages. |
