摘要: |
目的 探讨补阳还五汤促进EPCs修复损伤血管内皮的作用,从归巢环节阐释其作用机制,为中医药防治心脑血管疾病提供参考。方法 以补阳还五汤灌胃及尾静脉输注EPCs作用于内皮损伤的模型大鼠,从血管内皮形态、EPCs归巢等方面评价内皮损伤修复情况,从整合素αvβ3和αvβ5介导的SDF-1/CXCR-4信号通路了解该方促进EPCs归巢的机制。结果 与单用EPC组(E组)和单用药物组(b组)比较,补阳还五汤联合EPC组(B组)αvβ3表达显著增加(P<0.05),血管SDF-1表达显著增高(P<0.01)。结论 补阳还五汤能促进EPC修复损伤的血管内皮,其作用机制可能与该方能上调整合素αvβ3介导的SDF-1信号通路相关蛋白,促进EPCs的归巢有关。 |

Objective To investigate the role of Buyang Huanwu decoction (BYHWD) in promoting endothelial progenitor cell (EPC)-induced recovery of injuried endothelial cells, and explain the mechanism from EPCs homing. Methods The endothelial injuried rats were given gavage with BYHWD and vena caudalis injection of EPC. The repaired situation of injuried endothelium was observed. The effect of BYHWD on EPC homing was analyzed from integrin αvβ3 and integrin αvβ5-mediated SDF-1/CXCR-4 signaling pathway. Results Compared with group EPC (group E) and BYHED group (group b), the protein expression of vascular αvβ3 in BYHWD combined with EPC (group B) increased more significantly (P<0.05), vascular SDF-1 expression increased significantly (P<0.01). Conclusion BYHED could promote EPC repairing damaged endothelium, the mechanism may be related to promote EPCs homing through upregulating αvβ3-mediated SDF-1signaling pathway. |
