三七总皂苷预处理对大鼠肾缺血再灌注损伤的保护作用及机制研究 |

摘要: |
目的 探讨三七总皂苷对大鼠肾缺血再灌注损伤(renal ischemia reperfusion injury,RIRI)的保护作用,并研究其对氧化应激及炎症级联反应的影响。方法 将40只雄性SD大鼠随机均分为假手术组,模型组,三七总皂苷低、中、高剂量[40、80、160 mg/(kg·d)]预处理组,分别予以相应干预,连用5 d。在最后1次给药1 h后采用切除右肾、夹闭左肾动脉45 min后再灌注3 h的方法建立RIRI模型。五组均于再灌注3 h后经腹主动脉采血5 mL,检测血清中尿素氮(BUN)、血肌酐(SCr)、丙二醛(MDA)含量及超氧化物歧化酶(SOD)活性;取肾组织常规切片和HE染色,光镜下观察肾组织病理学改变;免疫组化法检测细胞间黏附因子-1(ICAM-1)、核因子-κB p65(NF-κB p65)蛋白表达。结果 与假手术组比较,模型组的SCr、BUN、MDA含量明显增高,SOD活性降低,光镜下可见肾小管损伤明显,免疫组化显示ICAM-1、NF-κB p65蛋白表达明显增高,差异有统计学意义(P<0.01);与模型组比较,三七总皂苷各剂量组的SCr、BUN、MDA水平明显降低,SOD活性增高,肾组织病理学改变明显减轻,ICAM-1、NF-κB p65表达显著降低,差异有统计学意义(P<0.01);三七总皂苷中、高剂量组组间比较,各指标差异均无统计学意义(P>0.05)。结论 三七总皂苷可通过减轻氧化应激和炎性损伤,有效改善大鼠肾缺血再灌注损伤。 |

Abstract: |
Objective To investigate the protective effect and its mechanism of panax notoginseng saponins (PNS) on renal ischemia reperfusion injury (RIRI) in rats.Methods 40 male SD rats were randomly divided into the sham-operation group,model group,low dose,medium dose,high dose[40,80,160 mg/(kg·d)]PNS groups.All rats were administered for 5 d,respectively.After ending treatment for 1 h,the RIRI model was established.After reperfusion for 3 h,the levels of urine nitrogen (BUN),secrumcreatinine (SCr),the content of malondialdehyde (MDA) and the activity of superoxide dismutase (SOD) in serum were measured.The pathological changes of kidney tissue were investigated by optical microscopy and hematoxylin-eosin (HE) staining techniques.The levels of intercellular cell adhesion molecule-1(ICAM-1) and nuclear factor kappa B p65(NF-κB p65) in renal tissue were evaluated by immunohistochemical staining.Results Compared with sham-operation group,the levels of serum Cr,BUN,MDA and renal tissue ICAM-1,NF-κB p65 increased in model group,the injury of renal tubule was obvious,SOD decreased,the differences were statistically significant (P<0.01).Compared with model group,the levels of serum Cr,BUN,MDA and renal tissue ICAM-1,NF-κB p65 in PNS groups decreased,pathological injury of kidney tissue relieved,while SOD increased,the differences were statistically significant (P<0.01).The indicators between PNS groups were not statistically significant (P>0.05).Conclusion PNS can improve the RIRI in rats through decreasing oxidative stress and inflammation. |
