摘要: |
目的 观察人参皂苷(Ginsenoside,GSS)对小鼠成骨细胞增殖活性的影响,并探讨其作用机制。方法 采用MC3T3-E1小鼠成骨细胞系, 加入不同浓度的GSS (3.125~50 μg/mL) 培养后,MTT法检测GSS对成骨细胞增殖的影响,荧光定量RT-PCR法检测不同浓度各组细胞内p21和p27 mRNA的表达水平,并采用Western-blot检测细胞周期相关蛋白表达。结果 与空白对照组相比,3.125~50 μg/mL的GSS在24、48 h时均能够显著促进成骨细胞的增殖(P<0.01或0.05),其作用具有时间依赖性和浓度依赖性。50、25 μg/mL的GSS能够显著抑制p21和p27的mRNA表达(P<0.01),而12.5 μg/mL的GSS对其表达无影响。50、25 μg/mL的GSS抑制了细胞周期抑制蛋白p21及p27表达水平并上调Cyclin D1表达(P<0.01或0.05)。结论 GSS能够通过上调Cyclin D1表达,抑制p21和p27表达,进而促进小鼠成骨细胞增殖. |

Effect of Ginsenosides on Promoting the Proliferation of Mouse Osteoblast and its Mechanisms |

Abstract: |
Objective To observe the effect of ginsenoside (GSS) on the proliferation of mouse osteoblast and explore the underlying mechanisms. Methods Mouse osteoblast MC3T3-E1 cells were incubated with different concentrations of GSS (3.125-50 μg/mL), and then the effect of GSS on proliferation was measured by MTT assay. The mRNA and protein levels of p21 and p27 were detected by real time RT-PCR and western blot respectively. Results Compared with the blank control group, different concentrations of GSS (3.125~50 μg/ml) all significantly promoted the proliferation of mouse osteoblast after treatment for 24 h and 48 h (P<0.01 or 0.05), the effects possess a dose- and time-dependent manner. 50 μg/mL and 25 μg/mL GSS significantly inhibited p21 and p27 mRNA levels (P<0.01), while 12.5 μg/mL GSS showed little effect on the its levels. 50 and 25 μg/mL GSS inhibited the expression of p21 and p27 protein and increased the level of Cyclin D1 (P<0.01 or P<0.05). Conclusion GSS can promote mouse osteoblast proliferation by increasing the Cyclin D1 expression and decreasing the p21 and p27 expression. |
