心痛泰颗粒对大鼠抗急性心肌缺血保护作用的实验研究 |

摘要: |
目的 探讨心痛泰颗粒对大鼠抗急性心肌缺血的保护作用的研究。方法 将大鼠随机分成6组,除假手术组10只外,余大鼠予以结扎大鼠冠状动脉左前降支造成大鼠急性心肌缺血损伤模型,成模大鼠随机分为模型组、心痛泰高、中、低剂量组及麝香保心丸组。分别给以蒸馏水、心痛泰低、中、高剂量及麝香保心丸进行灌胃2周后,观察3个时间点模型动物心电图S-T段变化;采用ELISA法检测血清肌酸激酶(CK)、乳酸脱氢酶(LDH)、丙二醛(MDA)和超氧化物歧化酶(SOD)含量;采用TTC染色测定心肌梗死面积;采用HE染色法观察心肌坏死组织病理变化。结果 与假手术组比较,模型组大鼠心电图S-T段抬高明显(P<0.01)提示模型复制成功,模型组血清CK、LDH和MDA的浓度明显升高(P<0.01),血清SOD浓度明显降低(P<0.01);与模型组比较,各给药组有拮抗大鼠急性心肌缺血心电图S-T段抬高的作用(P<0.05),且能明显降低血清CK、LDH活性和MDA浓度,升高血清SOD活性(P<0.01),明显缩小心肌梗死范围,保护心肌组织(P<0.01)。结论 心痛泰颗粒抗急性心肌缺血心肌损伤的效果显著,对急性心肌缺血损伤大鼠有明显的保护作用。 |

Abstract: |
Objective To investigate the protective effect of Xintaitong granule on the rats with acute myocardial ischemia. Methods The rats were randomly divided into six groups. Except for the10 rats in sham-operation group, the other rats were ligaturedleft anterior of coronary artery to establish the rat models of acute myocardial ischemia. The model rats were randomly divided into the model group, high-, medium-and low-dose of Xintongtai groups, and Shexiang Baoxin pill group, which were treatedseparately with distilled water, low-dose, medium-dose and high-dose of Xintongtai and Shexiang Baoxin pill by gavage for 2 weeks. Thechanges of ECG S-T segment at 3 time points of animal models were observed; Thecontentsof CK, LDH, MDA and SDA were detected by ELISA. The area of myocardial infarction was determined by TTC staining method; The pathological changes of myocardial necrosis tissue were observedby HE staining method. Results Compared with the sham-operation group, the S-T segment in model group rats was increased obviously(P<0.01); The serum CK, LDH and MDA content increased significantly(P<0.01), serum SOD content was significantly reduced(P<0.01). Compared with the model group, the given medicine groups could against ECG S-T segment elevation of acute myocardial ischemiain rat(P<0.05), they can significantly reduce the serum CK and LDH activity and MDA content, increase the serum SOD activity(P<0.01), significantly reduced the size of myocardial infarct size, protect myocardial tissues (P<0.01). Conclusion Xintongtai show obvious effect against acute myocardial ischemia myocardial injury and protective effect on rats with acute myocardial ischemia injury. |
