摘要: |
目的探讨湿痹通对大鼠急性痛风性关节炎的抗炎作用机制。方法将40 只大鼠随机分为空白组,模型
组,秋水仙碱组,湿痹通高、低剂量组共5 组,每组8 只。采用踝关节腔内注射尿酸钠的方法建立大鼠急性痛风性关节
炎模型,观察造模后8、72 h 各组大鼠炎症指数及功能障碍指数。检测各组大鼠血清白细胞介素-1(IL-1)、白细胞介素
(IL-1R)的含量。并测量各组大鼠单位面积关节软组织中炎症细胞因子白细胞介素-1β(IL-1β),IL-1R 的表达。结果
模型组炎症指数及功能障碍指数积分均比其它各组高,差异有统计学意义(P<0.05);模型组血清IL-1、IL-1R 含量及
关节软组织内IL-1β、IL-1R 的表达较其余各组增高,差异有统计学意义(P<0.05);其中关节软组织内IL-1β、IL-1R 的
表达湿痹通高剂量组比低剂量组降低,差异有统计学意义(P<0.05)。结论湿痹通能改善动物模型急性痛风性关节炎
的炎症反应,其作用机制可能是通过阻断血清及关节软组织内IL-1 和IL-1R 的表达。 |

Anti-inflammatory mechanism of Shibitong priscriptionon the acute gouty arthritis rats |

Abstract: |
Objective To observe the anti-inflammtory mechanism of Shibitong prscription on
rats with acute gouty arthritis. Methods 40 SPF Wistar rats were randomly divided into 5 groups
as follows: blank group, model group, colchicine group, Shibitong high -dose group and
low-dose group, 8 rats in each group. Rat model of acute gouty arthritis was replicated by intraarticular
injection of monosodium urate crystal suspension inside the ankle joint. Inflammation index
and dysfunction index were observed after modeled 8 h and 72 h. IL-1β and IL-1R expression
levels in serum and joint soft tissues were detected. Results The inflammation index
and dysfunction index, expression levels of IL-1β and IL-1R in serum and joint soft tissues of
the model group were higher than those of other groups (P<0.05), and expression levels of IL-1β
and IL-1R in joint soft tissues of the Shibitong high-dose group were higher than those of Shititong
low-dose group (P<0.05). Conclusion The inflammatory effects in rat with acute gouty arthritis
can be improvement by treating Shibitong priscription and the Shibitong effects may be related
with the decreased expression levels of IL-1β and IL-1R in serum and joint soft tissues. |
