| 摘要: |
| 目的:建立柱前衍生高效液相色谱法(HPLC)同时测定阿胶中17种水解氨基酸含量的方法。方法用6 mol/L盐酸溶液水解阿胶中的氨基酸,以异硫氰酸苯酯为柱前衍生试剂;用Hypersil BDS C18柱(200 mm×4.6 mm,5μm),梯度洗脱,流速为1.0 mL/min,检测波长为254 nm,柱温为30℃。结果 17种氨基酸在60 min内均可得到很好的分离,回收率为97.4%～100.1%,RSD为1.6%～3.4%。结论建立的方法可靠,可用于阿胶中17种水解氨基酸的含量测定。 |

| Abstract: |
| Objective To determine the contents of 17 kinds of hydrolyzed amino acids in donkey-hide glue by HPLC with pre一〔column derivatization at the same time. Methods To extract amino acids by the method of hydrolysis in 6 mol/L HCl and use phenylisothiocyanate (PITC) as the derivating agent. The Hypersil BDS C,8 column (200 mmx4.6 mm, 5μm) was used. Under the condition of gradient elution, the flow rate was 1.0 mL/min, the UV detection wavelength was 254 nm, and the column temperature was 30 0C. Results 17 kinds of amino acids can be separated well in 60 min. The average recoveries were 97.4%一100.1% and RSD were 1.6%}3.4%. Conclusion The method is reliable for the determination of 17 kinds of amino acids in donkey-hide glue. |
