基于传统知识调查和网络药理学的土家药天珠散增药组方抗脑卒中效益评价研究 |

摘要: |
目的 构建土家族方剂天珠散增药组方并探讨其抗脑卒中的效益性。方法 围绕疾病脑卒中,首先通过区域性问卷调查方法确定候选增药范围,然后利用网络药理学方法对候选药物与疾病靶点进行分析,确定最佳新组方,再依据土家族的“主帮客信”配伍理论,对新方展开合理性评析,最后利用分子对接方法,对新方治疗脑卒中的机制进行初步探究。结果 土家医推荐与天珠散配伍抗脑卒中的药物中,八角枫、三角风、头晕草排名靠前;对比各候选药物预测靶点与疾病靶点网络拓扑参数,确定了天珠散加头晕草为新方最佳配伍,其中天麻为主药,延龄草为帮药,头晕草可兼为帮、客、信药;新方可通过参与一氧化氮生物合成过程调控、磷酸化的正调控、对含氧化合物的反应、神经元投射维持等生物过程,对脑卒中疾病症状进行干预;分子对接结果显示,该组方成分26-chloro-26-deoxycryptogenin、β-ecdysterone、ecdysterone、gastrodin、4-hydroxycinnamic acid分别与蛋白APP、IL-6、MTOR、JAK2、TLR4具有良好的结合活性。结论 天珠散配伍头晕草具有合理性与效益性,其或可增加原方天珠散抗脑卒中效果。 |

Objective To construct Tujia medicine Tianzhusan added medicine prescription and explored its effectiveness in anti stroke. Methods Focusing on stroke, firstly, the scope of candidate herbals was determined by regional questionnaire survey, and network pharmacology was used to analyze the candidate herbals and disease targets to determine the best new prescription. Then, according to the compatibility theory "Zhu Bang Ke Xin" of Tujia doctors, the new prescription was rationally evaluated. Molecular docking technology was used to explore the mechanism of the new prescription in the treatment of stroke preliminarily.Results Among the herbals recommended by Tujia doctors for the treatment of stroke with Tianzhusan, Bajiaofeng (Alangium Chinense), Sanjiaofeng (Hedera Nepalensis) and Touyuncao (Geum Japonicum Thunb) were top ranked. By comparing the topological parameters of each candidate herbal prediction targets and disease targets network, it was determined that Tianzhusan plus Touyuncao (Geum Japonicum Thunb) was the best compatibility of the new prescription, in which Tianma (Gastrodiae Rhizoma) was the “Zhu” herbal, Yanlingcao (Trillium Tschonoskii) was the “Bang” herbal, and Touyuncao (Geum Japonicum Thunb) might act as the “Bang”, “Ke” and “Xin” herbal at the same time. The new prescription might intervene the symptoms of stroke by participating in the regulation of nitric oxide biosynthesis, the positive regulation of phosphorylation, the response to oxygenates, the maintenance of neuronal projection and other biological processes. The results of molecular docking showed that the constituents of 26-chloro-26-deoxycryptogenin, β-ecdysterone, ecdysterone, gastrodin and 4-Hydroxycinnamic acid had good binding activities with proteins APP, IL6, mTOR, JAK2 and TLR4 respectively. Conclusion Tianzhusan combined with Touyuncao (Geum Japonicum Thunb) is reasonable and effective, which may increase the effect of the original prescription Tianzhusan in the treatment of stroke. |
