| 目的 探究过山枫(Celastrus aculeatus)果实的化学成分及细胞毒活性。方法 运用多种色谱分离技术对过山枫果实醇提物进行分离纯化,借助核磁共振波谱和质谱技术鉴定所得化合物的结构。同时,采用MTT法检测倍半萜类化合物对急性粒细胞白血病细胞HL-60、人宫颈癌细胞HeLa和人肝癌细胞BEL-7402的细胞毒活性。结果 从过山枫果实中分离得到17个化合物,包含12个β-二氢沉香呋喃型倍半萜类化合物:1β,6α,8β,13-四乙酰氧基-9β-苯甲酰氧基-β-二氢沉香呋喃(1)、Ejap-4(2)、Chiapen C(3)、Ejap-3(4)、Ejap-2(5)、1β,6α,13-三乙酰氧基-9α-苯甲酰氧基-β-二氢沉香呋喃(6)、Ejap-10(7)、Celaspene A(8)、6α-乙酰氧基-1β,9β-二苯甲酰氧基-8β-羟基-β-二氢沉香呋喃(9)、1β,8α,13-三乙酰氧基-9β-苯甲酰氧基-β-二氢沉香呋喃(10)、Angulatueoid B(11)、1β,8α-二乙酰氧基-9α-苯甲酰氧基-12β-O-烟酰基-β-二氢沉香呋喃(12)及3个三萜类化合物β-香树脂醇(13)、11-甲氧基-3-乙酰氧基齐墩果烷(14)、11α-甲氧基-β-香树脂醇(15)和2个甾体类化合物β-谷甾醇(16)、胡萝卜苷(17)。除化合物13、16、17外,其余化合物均为首次从该植物中分离得到。与阳性药依托泊苷相比,所测试化合物对HL-60、HeLa、BEL-7402肿瘤细胞的细胞毒活性均较弱。结论 过山枫果实含有丰富的β-二氢沉香呋喃型倍半萜及三萜类化合物。尽管分离的倍半萜类化合物对肿瘤细胞未表现明显细胞毒性,但本研究丰富了过山枫果实的化学物质基础,为其药用部位的拓展提供了科学依据。 |

| Objective To investigate the chemical constituents in the fruits of Celastrus aculeatus and evaluate their cytotoxic activities. Methods Various chromatographic separation techniques were employed to isolate and purify the ethanol extract of Celastrus aculeatus fruits. The structures of obtained compounds were identified through nuclear magnetic resonance spectroscopy and mass spectrometry. The MTT assay was used to test the cytotoxic activities of isolated sesquiterpenoids against acute myeloid leukemia HL-60 cells, human cervical cancer HeLa cells, and human hepatocellular carcinoma BEL-7402 cells. Results A total of 17 compounds were isolated from the fruits of Celastrus aculeatus, including 12 dihydro-β-agarofuran-type sesquiterpenoids: (1) 1β, 6α, 8β, 13-tetraacetoxy-9β-benzoyloxy-β-dihydroagarofuran, (2) Ejap-4, (3) Chiapen C, (4) Ejap-3, (5) Ejap-2, (6) 1β, 6α, 13-triacetoxy-9α-benzoyloxy-β-dihydroagarofuran, (7) Ejap-10, (8) Celaspene A, (9) 6α-acetoxy-1β, 9β-dibenzoyloxy-8β-hydroxy-β-dihydroagarofuran, (10) 1β, 8α, 13-triacetoxy-9β-benzoyloxy-β-dihydroagarofuran, (11) Angulatueoid B, (12) 1β, 8α-diacetoxy-9α-benzoyloxy-12β-O-nicotinoyl-β-dihydroagarofuran; three triterpenoids: (13) β-amyrin, (14) 11-methoxy-3-acetoxyoleanane, (15) 11α-methoxy-β-amyrin; and two steroidal compounds: (16) β-sitosterol, (17) daucosterol. Except for compounds 13, 16, and 17, all other compounds were isolated from this plant for the first time. Compared with the positive control drug etoposide, the tested compounds exhibited very weak cytotoxic activities against the above tumor cells. Conclusion The fruits of Celastrus aculeatus contain abundant dihydro-β-agarofuran-type sesquiterpenoids and triterpenoids. Although the isolated sesquiterpenoids did not exhibit significant cytotoxic activity against the tumor cells, this study enriches the chemical constituents of this fruit and provides scientific evidence for further development of its medicinal parts. |
