| 基于VEGF-PI3K/Akt通路探究电针阳陵泉对缺血性脑卒中模型神经功能的影响 |

| 摘要: |
| 目的 探究电针对缺血性脑卒中大鼠神经功能的影响及潜在机制。方法 随机选择9只健康大鼠作为对照组,其余31只大鼠建立右侧颈总动脉血栓形成模型。将造模成功的大鼠随机分为模型组、尼莫地平组(腹腔注射10 mg/kg尼莫地平)和电针组(电针双侧阳陵泉),每组9只。采用Zea-Longa评分法评估大鼠神经功能缺损情况;水迷宫试验评估大鼠认知功能;TTC染色观察大鼠脑梗死面积;HE染色、Nissl染色观察大鼠海马组织病理损伤情况;Western blot检测海马组织血管内皮生长因子(VEGF)、血管内皮生长因子受体2(VEGFR2)、磷酸化磷脂酰肌醇3激酶(p-PI3K)、磷酸化蛋白激酶B(p-Akt)、核因子κB(NF-κB)蛋白表达水平。结果 与对照组相比,模型组大鼠神经功能评分增加(P<0.05),逃避潜伏期延长(P<0.05),目标象限停留时间缩短(P<0.05),穿越平台次数减少(P<0.05),脑梗死面积增加(P<0.05),神经元数量减少(P<0.05),VEGF、VEGFR2、p-PI3K、p-Akt、NF-κB蛋白表达水平均降低(P<0.05)。与模型组相比,尼莫地平组和电针组大鼠神经功能评分降低(P<0.05),逃避潜伏期缩短(P<0.05),目标象限停留时间延长(P<0.05),穿越平台次数增加(P<0.05),脑梗死面积减少(P<0.05),神经元数量增加(P<0.05),VEGF、VEGFR2、p-PI3K、p-Akt、NF-κB蛋白表达水平均升高(P<0.05)。结论 电针阳陵泉可能通过激活VEGF-PI3K/Akt信号通路,改善缺血性脑卒中大鼠神经病理损伤和认知功能障碍,从而减轻其神经功能损伤。 |

| Objective To investigate the effects of electroacupuncture on neurological function in rats with ischemic stroke and its potential mechanisms. Methods Nine healthy rats were randomly selected as the control group, and the remaining 31 rats were used to establish a model of right common carotid artery thrombosis. The successfully modeled rats were randomly divided into model group, nimodipine group (intraperitoneal injection with 10 mg/kg nimodipine), and electroacupuncture group [electroacupuncture at the bilateral Yanglingquan (GB34)], with 9 rats in each group. Zea-Longa score was used to evaluate the rat nerve function defect; Water maze test was conducted to evaluate cognitive function; Triphenyltetrazolium chloride (TTC) staining was used to observe the infarction area; Hematoxylin and eosin (HE) and Nissl staining were used to observe the pathological injury of rat hippocampus; Western blot was used to detect the protein expression levels of vascular endothelial growth factor (VEGF), vascular endothelial growth factor receptor 2 (VEGFR2), phosphorylated phosphatidylinositol 3-kinase (p-PI3K), phosphorylated protein kinase B (p-Akt), and nuclear factor-κB (NF-κB) in hippocampal tissues. Results Compared with the control group, the model group showed a significant increase in neurological deficit scores (P<0.05), prolonged escape latency (P<0.05), shortened time spent in the target quadrant (P<0.05), reduced number of platform crossings (P<0.05), increased cerebral infarct area (P<0.05), decreased number of neurons (P<0.05), and lower protein expression levels of VEGF, VEGFR2, p-PI3K, p-Akt, and NF-κB (P<0.05). Compared with the model group, the nimodipine group and the electroacupuncture group exhibited decreased neurological deficit scores (P<0.05), shortened escape latency (P<0.05), prolonged time spent in the target quadrant (P<0.05), increased number of platform crossings (P<0.05), reduced cerebral infarct area (P<0.05), increased number of neurons (P<0.05), and elevated protein expression levels of VEGF, VEGFR2, p-PI3K, p-Akt, and NF-κB (P<0.05). Conclusion Electroacupuncture at Yanglingquan (GB34) may alleviate neuropathological damage and improve cognitive dysfunction in rats with ischemic stroke by activating VEGF-PI3K/Akt signaling pathway. |
