| 目的 探究背部推法对慢性疲劳综合征（CFS）大鼠疲劳及认知功能、血管内皮细胞的影响。方法 将24只SD大鼠随机分为空白组（n=6）和造模组（n=18）。空白组常规饲养，造模组选用强迫负重游泳联合慢性应激刺激制备CFS大鼠模型，造模21 d。造模成功后将造模组随机分为模型组、推法组和药物组，每组6只。推法组在造模结束后给予背部推法，20 min/次；药物组给予人参皂苷水溶液灌胃，1 mL/次。两组均为1次/d，干预14 d。实验期间，记录各组大鼠体质量、一般情况半定量评分。干预结束后，记录旷场实验运动总路程及中心区域路程、Morris水迷宫逃避潜伏期及穿越原平台次数；收集大鼠血清及海马组织，采用荧光探针检测海马组织活性氧（ROS）含量，透射电镜观察海马组织血管内皮细胞超微结构，ELISA检测血清中血管紧张素Ⅱ（AngⅡ）、内皮素1（ET-1）、去甲肾上腺素（NE）含量。结果 （1）造模结束后，模型组体质量显著低于空白组（P<0.01），一般情况半定量评分显著高于空白组（P<0.01）；干预后，推法组和药物组体质量显著高于模型组（P<0.01），一般情况半定量评分显著低于模型组（P<0.01）。（2）与空白组比较，模型组旷场实验运动总路程及中心区域路程、Morris水迷宫实验穿越原平台次数均减少（P<0.01，P<0.05），逃脱潜伏期显著延长（P<0.01）；与模型组比较，推法组和药物组旷场实验运动总路程及中心区域路程、Morris水迷宫实验穿越原平台次数均增加（P<0.01，P<0.05），逃脱潜伏期显著缩短（P<0.01）。（3）超微结构显示，模型组血管内皮细胞结构破坏肿胀，推法组海马血管内皮细胞肿胀明显减轻。（4）与空白组比较，模型组海马组织ROS含量及血清AngII、ET-1、NE含量均显著升高（P<0.01）；与模型组比较，推法组和药物组上述指标含量均降低（P<0.05，P<0.01）。结论 背部推法能够降低CFS大鼠海马组织ROS，改善海马组织血管内皮细胞结构及功能，从而缓解疲劳及认知功能障碍。 |

| Effects of back pushing manipulation on the structure and function of hippocampal vascular endothelial cells in rats with chronic fatigue syndrome |

| Objective To investigate the effects of back pushing manipulation on fatigue and cognitive functions, as well as vascular endothelial cells, in rats with chronic fatigue syndrome (CFS). Methods A total of 24 SD rats were randomly assigned into blank group (n=6) and modeling group (n=18). The rats in blank group received routine feeding, while in the modeling group, the CFS rat model was established by forced weight-bearing swimming combined with chronic stress stimulation for 21 days. After successful modeling, the modeling group was further assigned into model, pushing manipulation, and drug groups, with six rats in each group. The pushing manipulation group received a 20-minute back pushing manipulation, while the drug group received 1 ml of ginsenoside aqueous solutian by gavage. All interventions were administered once daily for 14 consecutive days. Body weight, semi-quantitative scores of general condition were recorded during the experiment. The total distance and central area distance in the open field test, as well as the escape latency and number of crossing the original platform in the Morris water maze test were recorded after the intervention. Rat serum and hippocampal tissue were collected. The content of reactive oxygen species (ROS) in the hippocampal tissue was checked by fluorescent probe. The ultrastructure of hippocampal vascular endothelial cells was observed under transmission electron microscopy. Serum levels of angiotensin Ⅱ (Ang Ⅱ), endothelin-1 (ET-1), and norepinephrine (NE) were determined by ELISA. Results (1) Compared with the blank group, the body weight of the model group was significantly lower (P<0.01), while the semi-quantitative scores of general condition were significantly higher (P<0.01) after modeling; compared with the model group, the body weight of the pushing manipulation group and drug group was significantly higher (P<0.01), while the semi-quantitative scores of general condition were significantly lower (P<0.01) after intervention. (2) Compared with the blank group, the model group showed decreases in the total distance and the central area distance in the open field test, and fewer times of crossing the original platform in the Morris water maze test (P<0.01, P<0.05), along with a significant prolongation of escape latency (P<0.01); compared with the model group, the pushing manipulation group and the drug group showed significant increases in the total distance and the central area distance in the open field test, and more times of crossing the original platform in the Morris water maze test (P<0.01, P<0.05), along with a significant shortening of escape latency (P<0.01). (3) Ultrastructural observation showed structural damage and swelling of vascular endothelial cells in the model group, while the swelling of hippocampal vascular endothelial cells was obviously alleviated in the pushing manipulation group. (4) Compared with the blank group, the model group exhibited significantly higher content of ROS in the hippocampal tissue and serum content of Ang II, ET-1, and NE (P<0.01); compared with the model group, all the aforementioned indicators in the pushing manipulation group and the drug group decreased (P<0.05, P<0.01). Conclusion Back pushing manipulation can reduce the content of ROS in the hippocampal tissue and improve the structure and function of hippocampal vascular endothelial cells, thereby alleviating fatigue and cognitive dysfunction in CFS rats. |
