| 疏肝平喘方抑制Sirt6表达对哮喘小鼠Th17/Treg平衡的影响 |

| 摘要: |
| 目的 研究疏肝平喘方抑制Sirtuin6(Sirt6)表达对哮喘小鼠辅助性T细胞17(Th17)细胞和调节性T细胞(Treg)平衡的影响。方法 雄性BALB/c小鼠随机分为对照组、模型组(哮喘造模)、疏肝平喘组(哮喘造模+5.33 g/mL疏肝平喘方灌胃)、地塞米松组(哮喘造模+0.075 mg/mL地塞米松灌胃)。治疗1周后,通过HE染色和Masson染色观察肺组织病理改变及胶原沉积,采用整体体积描记法检测气道Penh值,ELISA检测血清和肺组织中白细胞介素(IL)-17、IL-10的水平,免疫组化染色检测肺组织中Sirt6的积分光密度(IOD)值,Western blot检测肺组织Sirt6、视黄酸相关孤儿受体γt(RORγt)、叉头框蛋白P3(FoxP3)的蛋白表达水平。结果 与对照组比较,模型组的肺组织可见肺泡壁增厚、平滑肌细胞增生、炎症细胞浸润等病理改变,胶原堆积增多;气道Penh值、血清及肺组织IL-17水平、肺组织中Sirt6的IOD值、肺组织Sirt6及RORγt的蛋白表达水平升高(P<0.05),血清及肺组织IL-10水平、肺组织FoxP3表达水平降低(P<0.05)。与模型组比较,疏肝平喘组和地塞米松组的肺组织病理改变减轻、胶原沉积减少;气道Penh值、血清及肺组织IL-17水平、肺组织Sirt6的IOD值、肺组织Sirt6及RORγt的蛋白表达水平降低(P<0.05),血清及肺组织IL-10水平、肺组织FoxP3表达水平升高(P<0.05)。结论 疏肝平喘方可改善哮喘小鼠的气道功能、肺组织病理改变、胶原堆积及Th17/Treg失衡,抑制Sirt6表达是疏肝平喘方发挥上述改善作用的可能机制。 |

| Effects of Shugan Pingchuan Formula on the Th17/Treg balance in asthmatic mice by inhibiting Sirt6 expression |

| Objective To investigate the effects of the Shugan Pingchuan Formula (SGPCF) on the balance of helper T cell 17 (Th17) and regulatory T cell (Treg) in asthmatic mice by inhibiting the expression of Sirtuin6 (Sirt6). Methods Male BALB/c mice were randomly divided into control group, asthma model group (asthma modeling), SGPCF group (asthma modeling+5.33 g/mL SGPCF by intragastric administration), and dexamethasone group (asthma modeling+0.075 mg/mL dexamethasone by intragastric administration). After one week of treatment, the pathological changes of lung tissue and collagen deposition were observed by HE staining and Masson staining, respectively. The Penh value of the airway was checked by total volume tracing. The levels of interleukin (IL)-17 and IL-10 in serum and lung tissue were measured by ELISA. The integral optical density (IOD) value of Sirt6 in lung tissue was determined by immunohistochemical staining. The protein expression levels of Sirt6, retinoic acid receptor-related orphan receptor γt (RORγt), and forkhead box protein P3 (FoxP3) in lung tissue were assessed by Western blot. Results Compared with the control group, the asthma model group showed pathological changes in lung tissue, including alveolar wall thickening, proliferation of smooth muscle cells, inflammatory cell infiltration, and increased collagen deposition. The airway Penh value, IL-17 levels in serum and lung tissue, IOD value of Sirt6 in lung tissue, and the protein expression levels of Sirt6 and RORγt were significantly increased (P<0.05). IL-10 levels in serum and lung tissue, as well as FoxP3 expression in lung tissue, were significantly decreased (P<0.05). Compared with the asthma model group, both the SGPCF and the dexamethasone groups exhibited alleviated lung pathological changes and reduced collagen deposition. The airway Penh value, IL-17 levels in serum and lung tissue, IOD value of Sirt6, and protein expression levels of Sirt6 and RORγt in lung tissue were significantly decreased (P<0.05). IL-10 levels in serum and lung tissue, as well as the expression level of FoxP3 in lung tissue were significantly increased (P<0.05). Conclusion The SGPCF can improve airway function, and alleviate pathological changes, collagen deposition, and Th17/Treg imbalance in lung tissue of asthmatic mice. Downregulation of Sirt6 expression is the possible mechanism by which the SGPCF exerts the above effects. |
