| 不同方式注射垂体后叶素对大鼠心肌缺血模型影响的研究 |

| 摘要: |
| 目的 比较腹腔注射、尾静脉注射以及股静脉注射3种注射方式注射垂体后叶素诱导急性心肌缺血大鼠模型的差异。方法 32只SD雄性大鼠随机分为4组：正常组、腹腔注射组、尾静脉注射组和股静脉注射组（n=8）。正常组注射生理盐水，其余3组分别采用不同注射方式注射垂体后叶素诱导心肌缺血。通过观察心电图上J点位置，血清血清肌钙蛋白I（cardiac troponin I,cTn-I）、肌酸激酶同工酶MB（creatine kinase isoenzyme-MB,CK-MB）、乳酸脱氢酶（lactate dehydrogenase,LDH）含量变化和HE染色心肌组织病理变化来对比模型差异。结果 （1）与正常组相比，各模型组均出现J点位移变化，差异有统计学意义（P＜0.01）；股静脉注射组J点位移变化值大于腹腔注射组和尾静脉注射组。重复测量方差分析结果显示：各组大鼠心电图J点位移值在不同时间之间有差异（P<0.001）；时间与组间有交互效应，不同注射方式之间有差异（P<0.001）。（2）与正常组相比，各模型组血清cTn-I、CK-MB、LDH含量均升高，差异有统计学意义（P＜0.01）；尾静脉注射组与腹腔注射组效应相当；股静脉注射组血清cTn-I、CK-MB含量高于腹腔注射组，血清LDH含量高于腹腔注射组和尾静脉注射组（P＜0.05）。（3）与正常组相比，各模型组心肌组织均出现心肌损伤性病理改变，以股静脉注射组病理表现最明显。结论 股静脉注射垂体后叶素建立的急性心肌缺血大鼠动物模型病理和临床表现比腹腔注射和尾静脉注射建立的模型更明显。 |

| Objective To compare the difference of acute myocardial ischemia models induced by pituitrin in rats by intraperitoneal injection, caudal vein injection and femoral vein injection. Methods A total of 32 male SD rats were randomly divided into four groups:normal group, intraperitoneal injection group, caudal vein injection group and femoral vein injection group (n=8). The normal group was injected with normal saline, and the other three groups were injected with pituitrin in different ways to induce myocardial ischemia. The difference of the model was compared by observing the location of J-point on electrocardiogram, the changes of serum cardiac troponin I (cTn-I), creatine kinase isoenzyme-MB (CK-MB), lactate dehydrogenase (LDH) and the pathological changes of myocardial tissue stained by HE. Results (1) Compared with the normal group, there were J-point displacement changes in each model group, and the difference was statistically significant (P<0.01), and the J-point displacement of the femoral vein injection group was larger than that of the intraperitoneal injection group and the caudal vein injection group. The results of repeated measures analysis of variance showed that the displacement of J-point in electrocardiogram of each group was different at different time (P<0.001); there was an interactive effect between time and groups, and there was a difference between different injection methods (P<0.001). (2) Compared with the normal group, the levels of serum cTn-I, CK-MB and LDH in the model groups were significantly increased, the difference was statistically significant (P<0.01); the effect of caudal vein injection group was similar to that of intraperitoneal injection group; the levels of serum cTn-I and CK-MB in the femoral vein injection group were higher than those in the intraperitoneal injection group, and the level of serum LDH in the femoral vein injection group was higher than that in the intraperitoneal injection group and the caudal vein injection group (P<0.05). (3) Compared with the normal group, myocardial injury pathological changes were found in all model groups, and the most obvious pathological changes were found in the femoral vein injection group. Conclusion The animal model of acute myocardial ischemia induced by injection of pituitrin via femoral vein injection has more obvious pathological and clinical manifestations than that induced by injection of pituitrin intraperitoneal injection or caudal vein injection. |
