| 摘要: |
| 目的 探究益气活血方对肝纤维化模型大鼠的干预效果及相关作用机制。方法 选取27只SD健康雄性大鼠，其中9只为正常组，其余18只建立肝纤维化模型，并随机分为模型组、益气活血方组，每组9只，益气活血方组大鼠使用4 mL益气活血方灌胃干预，正常组、模型组大鼠使用4 mL生理盐水灌胃干预，各组大鼠均连续干预8周。计算各组大鼠肝脏系数、脾脏系数、肝纤维化程度评分，全自动生化分析仪检测肝功能指标[天冬氨酸转氨酶（AST）、丙氨酸转氨酶（ALT）]、肝纤维化指标[透明质酸酶（HA）、Ⅲ型前胶原（PCⅢ）、层粘连蛋白（LN）]水平，酶联免疫吸附实验法检测炎症因子[白介素-3（IL-3）、肿瘤坏死因子-α（TNF-α））]、氧化应激反应指标[过氧化氢酶（CAT）、晚期氧化蛋白产物（AOPP）]水平，Western blot法检测TGF-β/Smads通路蛋白及线粒体凋亡通路蛋白相对表达量。结果 与正常组比较，其他两组大鼠肝脏系数、脾脏系数较高；与模型组比较，益气活血方组大鼠肝脏系数、脾脏系数、肝纤维化程度评分较低（P<0.05）。与正常组比较，其他两组大鼠AST、ALT、HA、PCⅢ、LN、IL-3、TNF-α、AOPP水平较高，CAT水平较低；与模型组比较，益气活血方组大鼠AST、ALT、HA、PCⅢ、LN、IL-3、TNF-α、AOPP水平较低，CAT水平较高（P<0.05）。与正常组比较，其他两组大鼠Smad3、Smad2、TGF-β1、Bcl-2相对表达量较高，Bax、Caspase-3相对表达量较低；与模型组比较，益气活血方组大鼠Smad3、Smad2、TGF-β1、Bcl-2相对表达量较低，Bax、Caspase-3相对表达量较高（P<0.05）。结论 使用益气活血方对肝纤维化模型大鼠进行干预，改善大鼠肝功能，减轻肝纤维化程度及炎症反应、氧化应激反应，其作用机制可能为益气活血方干预能够调控TGF-β/Smads通路及线粒体凋亡通路蛋白表达，从而起到肝脏保护作用。 |

| To Explore the Intervention Effect of Yiqi Huoxue Formula on Liver Fibrosis Model Rats Based on Mitochondrial Apoptosis Pathway |

| Objective To explore the intervention effect and related mechanism of Yiqi Huoxue Formula on liver fibrosis model rats. Methods 27 SD healthy male rats were selected, 9 of which were in the normal group, and the remaining 18 were established to model liver fibrosis, and were randomly divided into model group, Yiqi Huoxue Formula group, 9 rats in each group. Rats in Yiqi Huoxue Formula group were given 4 mL Yiqi Huoxue Granule intragastric intervention, while rats in normal group and model group were given 4 mL normal saline intragastric intervention, rats in each group were continuously intervened for 8 weeks. The scores of liver and spleen coefficient and liver fibrosis degree were calculated. The levels of liver function indexes[aspartate aminotransferase (AST), alanine aminotransferase (ALT)] and liver fibrosis indexes[hyaluronidase (HA), procollagen type Ⅲ (PCⅢ), laminin (LN)] were detected by automatic biochemical analyzer. The levels of inflammatory factors[interleukin (IL-3), tumor necrosis factor-alpha (TNF-α)] and oxidative stress indexes[catalase (CAT), advanced oxidation protein products (AOPP)] were detected by ELISA. The relative expression levels of TGF-β/Smads pathway protein and mitochondrial apoptosis pathway protein were detected by Western blot. Results Compared with normal group, liver coefficient and spleen coefficient of the other two groups were higher. Compared with model group, liver coefficient, spleen coefficient and liver fibrosis degree score of rats in Yiqi Huoxue Formula group were lower (P<0.05). Compared with the normal group, the levels of AST, ALT, HA, PCⅢ, LN, IL-3, TNF-α and AOPP in the other two groups were higher, and the level of CAT was lower. Compared with model group, the levels of AST, ALT, HA, PCⅢ, LN, IL-3, TNF-α and AOPP in Yiqi Huoxue Formula group were lower, and the level of CAT was higher (P<0.05). Compared with the normal group, the relative expression levels of Smad3, Smad2, TGF-β1 and Bcl-2 were higher in the other two groups, while the relative expression levels of Bax and Caspase-3 were lower in the other two groups. Compared with model group, the relative expression levels of Smad3, Smad2, TGF-β1 and Bcl-2 in Yiqi Huoxue Formula group were lower, while the relative expression levels of Bax and Caspase-3 were higher (P<0.05). Conclusion Yiqi Huoxue Formula was used to intervent liver fibrosis model rats can improve the rat liver function, reduce the degree of liver fibrosis and inflammation, oxidative stress, its mechanism may be that Yiqi Huoxue Formula intervention can control TGF-β/Smads pathway and mitochondrial apoptosis protein expression, thereby playing a protective role in liver. |
