参芪补肺方对慢性阻塞性肺疾病稳定期大鼠Nrf2和γ-GCS表达的影响 |

摘要: |
目的 探讨参芪补肺方对大鼠肺组织红系衍生核因子相关因子-2(nuclear factor erythroid 2-related factor 2,Nrf2)、γ-谷氨酰半胱氨酸合成酶(gamma-glutamylcysteines-ynthetase,γ-GCS)的mRNA及蛋白表达水平的影响。方法 将40只Wistar大鼠随机分为空白组、模型组、中药组、富露施组,每组各10只。除空白组外,其余3组均采用香烟烟雾熏制和经鼻给予脂多糖(lipopolysaccharides,LPS)制作慢性阻塞性肺疾病(chronic obstructive pulmonary disease,COPD)大鼠模型,且给予相应干预,连续灌胃30 d。于第38天对大鼠进行麻醉并提取肺组织,通过RT-PCR法检测Nrf2和γ-GCS的mRNA表达水平,通过免疫组化法、Western blot法检测Nrf2和γ-GCS蛋白表达水平。结果 中药组、富露施组大鼠Nrf2、γ-GCS的mRNA和蛋白表达均低于模型组,差异有统计学意义(P<0.05);中药组大鼠Nrf2、γ-GCS的mRNA和蛋白表达与富露施组组间比较差异无统计学意义(P>0.05),疗效相当。结论 参芪补肺方能够通过降低Nrf2、γ-GCS的mRNA及蛋白的表达,减轻COPD大鼠的氧化应激反应。 |

Effect of Shenqi Bufei Decoction on the Expression of Nrf2 and γ-GCS in Rats with Stable Chronic Obstructive Pulmonary Disease |

Abstract: |
Objective To investigate the effect of Shenqi Bufei Decoction on the mRNA and protein expression levels of nuclear factor erythroid 2-related factor 2 (Nrf2), Gamma-glutamylcysteine synthetase (γ-GCS) in rat lung tissue.Methods A total of 40 Wistar rats were equally and randomly divided into blank group, model group, traditional Chinese medicine group, and Fluimucil group. All rats except those in the blank group underwent cigarette smoke fumigation and nasal administration of lipopolysaccharide (LPS) to establish a rat model of chronic obstructive pulmonary disease (COPD), and received the corresponding intervention by gavage for 30 consecutive days. On the 38th day, the rats were given anesthesia and their lung tissues were collected. The mRNA expression levels of Nrf2 and γ-GCS were measured by RT-PCR, and the protein expression levels of Nrf2 and γ-GCS were measured by immunohistochemistry and Western blot.Results The mRNA and protein expression of Nrf2 and γ-GCS in the model group was significantly higher than that in the Fluimucil group and the traditional Chinese medicine group (P<0.05). There were no significant differences in the mRNA and protein expression of Nrf2 and γ-GCS between the traditional Chinese medicine group and the Fluimucil group (P>0.05).Conclusion Shenqi Bufei Decoction can reduce the mRNA and protein expression of Nrf2 and γ-GCS, so as to reduce the oxidative stress response of COPD rats. |
