雌激素受体α基因Xbal、PvuⅡ位点多态性与女性膝骨关节炎易感性关系的Meta分析 |

摘要: |
目的 探讨雌激素受体α(ER-α)基因Xbal、PvuⅡ位点多态性与女性膝骨关节炎(KOA)易感性的关系。方法 通过计算机检索Pubmed、EMBASE、CENTRAL、Web of science、Ovid、知网、维普和万方数据库,收集有关ER-α基因XbaI(A/G)或PvuⅡ(T/C)位点多态性与女性KOA易感性的研究。提取相关数据,计算OR值及95% CI,利用STATA 11软件行Meta分析。结果 共纳入7篇文献,9个病例-对照或队列研究,试验组2 194例,对照组2 959例。总体结果显示,XbaI、PvuⅡ位点多态性与女性KOA易感性均无显著相关(P>0.05)。亚组分析结果显示,XbaI位点多态性(G vs A;GG vs AA;GG+AG vs AA;GG vs AG+AA)可显著降低亚洲女性KOA的发病风险(P<0.01);PvuⅡ位点多态性(C vs T;CC vs TT;CC vs CT+TT)可显著增加亚洲女性KOA的发病风险(P<0.01)。结论 本研究提示ER-α基因Xbal、PvuⅡ位点多态性与女性KOA易感性存在种族差异,但本实验纳入样本量较少,结论仍需更多研究予以证实。 |

Association Between Estrogen Receptor Alpha Gene XbaI and PvuⅡ Polymorphisms and Susceptibility to Knee Osteoarthritis in Women: A Meta-analysis |

Abstract: |
Objective To investigate the association between estrogen receptor alpha (ER-α) gene XbaI and PvuⅡ polymorphisms and susceptibility to knee osteoarthritis (KOA) in women. Methods A literature search was performed in PubMed, EMBASE, CENTRAL, Web of Science, Ovid, CNKI, VIP, and Wangfang Data to collect the studies on the association between ER-α gene XbaI (A/G) and PvuⅡ (T/C) polymorphisms and susceptibility to KOA in women. The relevant data were extracted, and then odds ratio and 95% confidence interval were calculated. A meta-analysis was performed using STATA 11 software. Results A total of 9 case-control or cohort studies in seven publications involving 2194 KOA cases and 2959 controls were included in the meta-analysis. By pooling all the studies, either XbaI or PvuⅡ polymorphism was not significantly associated with KOA risk in women (P>0.05). In the subgroup analysis by ethnicity, XbaI polymorphism (G vs A; GG vs AA; GG+AG vs AA; GG vs AG+AA) was significantly associated with decreased KOA risk in Asian women (P<0.01); PvuⅡ polymorphism (C vs T; CC vs TT; CC vs CT+TT) was significantly associated with increased KOA risk in Asian women (P<0.01). Conclusion ER-α gene PvuⅡ and XbaI polymorphisms are found to be associated with susceptibility to KOA in Asian women only. However, the sample size of this analysis is small, and more studies are needed to confirm this conclusion. |
