摘要: |
目的 研究蛴螬提取物口服和滴眼两种途径对年龄相关性黄斑变性(age-related macular degeneration,AMD)模型干预机制。方法 采用光损伤方法建立干性AMD活体模型,分别予以蛴螬提取物口服和滴眼,用药4周后,观察其对各细胞层半胱氨酸天冬氨酸蛋白酶-3(cysteine aspastic acid-specific protease 3,Caspase-3)、核转录因子κB (nuclear factor-κB,NF-κB)、跨膜蛋白(Fas ligand,FasL)、肿瘤坏死因子α(tumor necrosis factor-α,TNF-α)表达的影响。结果 与空白组比较,模型组视网膜组织中的Caspase-3表达明显下降,FasL、TNF-α、NF-κB表达均增强(P<0.05),口服组和滴眼组组织中各项指标差异均无统计学意义(P>0.05);与模型组比较,口服组和滴眼组中Caspase-3表达明显升高,FasL、TNF-α、NF-κB表达明显降低;与口服组比较,滴眼组所有指标差异均无统计学意义(P>0.05)。结论 蛴螬提取物对感光细胞等细胞凋亡具有调节作用,表现为早期抑制,晚期促进。其调节作用可能是通过抑制或促进细胞凋亡关键因子Caspase-3表达实现的,同时通过抑制炎症相关因子FasL、TNF-α、NF-κB表达对慢性炎症具有抑制作用。 |

Abstract: |
Objective To study the interventional mechanism of oral administration and eye drops of the grub extract on the age-related macular degeneration (AMD) model. Methods A Dry AMD living model was established by light damage method. Models were given grubs extract oral administration and eye drops respectively. After 4 weeks, the expression of cysreine aspastic acid-specific protease 3 (Caspase-3), nuclear factor-κB (NF-κB), Fas ligand (FasL), tumor necrosis factor-α (TNF-α) was observed. Results Compared with the blank group, the expression of Caspase-3 in the retina tissue of model group decreased significantly, and the expression of FasL, TNF-α and NF-κB increased (P<0.05). Compared with the blank group, all indexes of the oral and eye drops groups had no significant difference (P>0.05). Compared with the model group, the expression of Caspase-3 increased significantly in the oral group and the eye drops group, and the expression of FasL, TNF-α and NF-κB decreased significantly. Compared with the oral group, there was no significant difference in all indicators in the eye drops group (P>0.05). Conclusion Grub extract has regulating action on photoreceptor cells apoptosis, such as performance for early suppression and lately acceleration. Its regulating effect is probably achieved by inhibiting or promoting the expression of Caspase-3. Grub extract shows inhibiting effect on chronic inflammation by inhibiting the expression of related inflammation factors, such as FasL, TNF-α, NF-κB. |
