强心安神汤对慢性心衰大鼠AngⅡ及AT1mRNA表达的影响 |

摘要: |
目的 研究强心安神汤对Wistar慢性心衰模型大鼠AngⅡ含量及AT1 mRNA表达的影响,探讨其可能的作用机制。方法 除空白组外,将Wistar大鼠造模后分为模型组、西药组、强心安神汤高剂量组及强心安神汤低剂量组,干预4周后,抽静脉血以免疫组化法测定大鼠血清中AngⅡ的含量;处死大鼠,提取心肌组织,以RT-PCR法检测心肌组织AT1 mRNA的表达。结果 强心安神汤组大鼠血清AngⅡ含量及AT1 mRNA表达与模型组及西药组比较,差异有统计学意义(P<0.05或P<0.01)。结论 强心安神汤治疗心衰的机制可能与其抑制AngⅡ引起的AT1 mRNA表达上调有关。 |

Abstract: |
Objective To study the effect of Qiangxin Anshen decoction on the expression of AngⅡand AT1 mRNA in the model of Wistar rats with chronic heart failure, and investigate its possible mechanism. Methods Except for the blank group, the model rats were randomly divided into model group, Western medicine group, high-dose Qiangxin Anshen decoction group and low-dose Qiangxin Anshen decoction group. After intervention treatment for 4 weeks, the content of AngⅡof serum in rats was measured. The rats were killed and the AT1 mRNA expression in myocardial tissues were detected. Results Compared with the model and Western medicine groups,the content of AngⅡ in the rats serum and AT1 mRNA expression of myocardial tissues in Qiangxin Anshen decoction group was statistically significant (P<0.05 or P<0.01), Conclusion The mechanism of Qiangxin Anshen decoction in treating heart failure may be related with the up-regulation of AT1 mRNA expression caused by restraining Ang Ⅱ. |
