回神颗粒对双转基因阿尔茨海默病小鼠海马区Aβ1-42表达的影响 |

摘要: |
目的 探讨在阿尔茨海默病(Alzheimer’s disease,AD)病理早期采用回神颗粒对转基因AD小鼠的治疗作用。方法 采用聚合酶链式反应(PCR)法鉴定淀粉样前体蛋白/早老素1(APP/PS1)双转基因AD小鼠的基因表型后,选取3月龄雄性转基因阳性PAD小鼠和同龄、同背景的C57BL/6J野生型雌性小鼠作为实验对象。应用Morris水迷宫中逃避潜伏期、穿越平台的次数来评价小鼠的学习记忆能力,苏木精-伊红(HE)染色法观察小鼠脑组织形态学的改变,免疫组化法检测小鼠海马区β淀粉样蛋白(β-amyloid protein,Aβ)1-42的表达水平。结果 回神颗粒治疗组小鼠学习记忆能力提高,与模型组比较,小鼠逃避潜伏期缩短,穿越平台次数增多(P<0.05);穿越平台次数与正常组比较差异无统计学意义(P>0.05);治疗组大脑海马区Aβ1-42表达阳性面积明显减少(P<0.05)。与正常组相比,模型组AD小鼠大脑海马区Aβ1-42表达阳性面积明显增高(P<0.05)。结论 回神颗粒可显著改善APP/PS1转基因AD小鼠的学习记忆能力,抑制Aβ1-42的过度表达、聚集。 |

Effects of Huishen Granule on Expression of Aβ1-42 in Hippocampus of Double-Transgenic AD Mice |

Abstract: |
Objective To explore the therapeutic effect of Huishen granule on early-stage Alzheimer's disease (AD) in transgenosis AD mice. Methods The genotyping of amyloid precursor protein/presenilin 1(APP/PS1) in double-transgenic AD mice were detected by PCR method. The 3-month male transgenic PAD mice and male C57BL/6J wild-type mice with same background were selected as experimental objectives.The ability of learning and memory of the rats were evaluated by escape latency and crossing times of Morris water maze.Themorphology changes in the brains of AD mice were observed by using Hematoxylin-Eosin(HE) staining method. β-amyloid protein 1-42 (Aβ1-42) in the area of hippocampus was detected by immunohistochemical method. Results Compared with the model group,the learning and memory ability in the Huishen granule treatmentgroup was increased, the escape latency was shorten and crossing times was increased (P<0.05); The crossing times in the treatment group were not significantly different from those in the normal group(P>0.05); The positive area of Aβ1-42 expression hippocampus in the treatment group were reduced(P<0.05). Compared with the model group,the positive area of Aβ1-42 expression in hippocampus of the model group were increasedobviously(P<0.05). Conclusion The Huishen granule could significantly improve the learning and memory ability in APP/PS1 double-transgenic AD mice and inhibit the over expression and accumulation of Aβ1-42. |
