摘要: |
〔摘要〕目的探讨大豆葺元对子宫内膜癌细胞增殖的影响及其对雌激素受体报告基因表达的影响、方法体
外培养子宫内膜癌细胞子宫内膜癌雌、孕激素受体阴性细胞系(HEC 1B),采用四甲基偶氮哩盐法(M1"P)检测不同
浓度大豆葺元对子宫内膜癌细胞增殖的影响,同时检测大豆葺元对雌激素应答原件(ERE)调控的雌激素受体。
(ERa)和雌激素受体(3 ( ER(3)的荧光素酶报告基因表达的影响〔结果大豆葺元对HEC-1B细胞体外增殖有明显
抑制作用,差异有统计学意义(P<0.01),其抑制作用有明显量效和时效性特征〔在浓度为20-80x 1 O}mol/L的大豆葺
元作用卜,报告基因1u。的表达较正常对照组明显提高,差异有统计学意义(P<0.05 ),在80x 10-`mol/L时到达最高
值;大豆葺元通过ER日介导的报告基因表达水平的升高程度强于ERa,差异有统计学意义(P<0.01)〔结论大豆葺
元可通过调节子宫内膜癌细胞ER。和ER日介导的报告基因的表达,调整ERa/ER日比例,从而抑制子宫内膜癌细

Abstract: |
(Abstract〕 Objective To study the effect of daidzein on the proliferation of endometri-
al carcinoma in uterus and its mechanism. Methods Endometrial carcinoma cell HEC一1B
were cultured in vitro, the influences of genintein in different doses on the proliferation of
HEC一1B cell were detected by MTT method, and the expression of luciferase reportor gene of
ERcx and ERpeffected by ERE were evaluated. Results Daidzein inhibited significantly the
proliferation of HEC一1B cells in vitro(P<0.01) and the effect was streghtened along with the in-
crease of the dose and the prolong of action time. Under the control of daidzein concentration
2080x10-bmol/L, the expression of reporter gene luc was increased compared with control group
(P<0.05) and reached the highest point in the concentration 80x10}mo1/L; the increase of二-
porter gene expression induced by ERp was stronger than that induced luc expression either
through ERcx (P<0.01). Conclusion Daidzein can inhabit the proliferation endometrial carcinoma
through regulating the expression of ERcx or ER(3, and regulating the balance of ERcx/ER(3. |
