| 摘要: |
| 目的 探讨益肾通癃胶囊对前列腺增生（benign prostatic hyperplasia，BPH）模型大鼠雌雄激素比及缺氧诱导因子1α（chypoxia-inducible factor-1α，HIF-1α）的影响。方法 对SD雄性大鼠皮下注射丙酸睾酮5 mg/（kg·d），连续4周。造模成功后将大鼠随机分为4组：正常组、模型组、中药对照组和实验组，每组7只。实验组大鼠灌服0.365 g/kg益肾通癃胶囊混悬液；中药对照组灌服0.183 g/kg癃闭舒胶囊混悬液；正常组及模型组给予同等容量的生理盐水灌胃，连续灌胃8周后，光镜下观察前列腺组织病理学变化；称取前列腺湿重并计算前列腺指数；检测大鼠血清E2、T、DHT水平及E2/T；测定前列腺组织中HIF-1α蛋白表达。结果 与模型组比较，实验组大鼠前列腺腺体轻微增生，少数呈乳头状增生，腺腔基本恢复正常。与模型组比较，实验组大鼠前列腺体积、前列腺湿重、前列腺指数及前列腺组织中的HIF-1α蛋白表达水平均降低（P<0.01）；大鼠血清中的DHT、E2、T水平降低，E2/T比值升高（P<0.01）。结论 益肾通癃胶囊可能通过降低BPH模型大鼠血清中的DHT、E2、T水平，升高E2/T比值，抑制HIF-1α的表达起治疗BPH的作用。 |

| Objective To investigate the effects of Yishen Tongzhi Capsules on the estrogen and androgen ratio in rat model of benign prostatic hyperplasia and the effects on hypoxia-inducible factor-1α(HIF-1α). Methods SD male rats were injected subcutaneously with testosterone propionate 5 mg/(kg·d) for 4 weeks. After successful modeling, the rats were randomly divided into 4 groups:a normal group, a model group, a Chinese medicine control group and an experimental group, with 7 rats in each group. Rats in the experimental group were administrated with 0.365 g/kg Yishen Tonglong Capsules Suspension; the Chinese medicine control group was administrated with 0.183 g/kg Longbishu Capsules Suspension; the normal group and the model group were given the same volume of saline. After 8 weeks of continuous intragastric administration, the histopathological changes of prostate were observed under light microscope; The prostate wet weight was measured and the prostate index was calculated; The rat serum E2, T, DHT levels, and E2/T were detected; HIF-1α protein expression in prostate tissue was measured. Results Compared with the model group, the prostate glands of the experimental group had a slight hyperplasia, and a few showed papillary hyperplasia. The glandular cavity returned to normal. Compared with the model group, rats in the experimental group had reduced prostate volume, wet prostate weight, prostate index, and HIF-1α protein expression levels in prostate tissue (P<0.01); DHT, E2, and T levels in rat serum were reduced. E2/T ratio increased (P<0.01). Conclusion Yishen Tonglong Capsules may treat BPH by reducing the levels of DHT, E2, T in the serum of BPH model rats, increasing the E2/T ratio and inhibiting the expression of HIF-1α. |
